BRS2 — Methylation & One-Carbon Metabolism

BRS2-3-7 - Phosphatidylcholine Formation

(PEMT methylation and Kennedy-pathway phosphatidylcholine synthesis)

1. Mission, Objective & Biological Context

Mission

Maintain phosphatidylcholine formation through PEMT methylation and the choline-based Kennedy pathway.

Intervention Dominance: Diet-Supported — Dietary Requirements

Conditional animal evidence concerns folate restriction and methyl-derived PC enrichment; it does not establish absolute PEMT flux, benefit above adequacy or cognitive benefit. Dietary prominence is supported within the studied animal context; it is not a head-to-head ranking against lifestyle or optimisation, a human intake target, or evidence of benefit from extra intake.

Overview

Cells form phosphatidylcholine through distinct routes: hepatic PEMT methylates phosphatidylethanolamine using SAMe, while the Kennedy pathway combines choline-derived phosphocholine with CTP and then diacylglycerol. Uridine can contribute upstream to nucleotide supply. These routes support membrane phospholipid provision; precursor availability does not establish that extra intake increases human membrane synthesis or improves cognition. [19] [20] [1]

  • Benefits: PC formation supplies a major membrane phospholipid. PEMT/choline-route loss in mice demonstrates the importance of maintaining hepatic PC provision; the new uridine evidence measures precursor formation, not a clinical benefit. [1] [19]
  • Implementation Notes: Choline and phosphatidylcholine can supply the existing methyl-donor provision route. Uridine adds a supported nucleotide-precursor route, with no established ordinary-food dose or human PC-flux benefit. [20] [21]
  • Biological Relevance: Only the PEMT branch directly couples methylation to membrane chemistry in the parent FM. Kennedy synthesis is a parallel PC-forming route; brain DHA delivery and incorporation remain separate downstream jobs. [1] [19]

2. Primary Biological Effects

Phosphatidylcholine formation through distinct PEMT and Kennedy routes; S-adenosylhomocysteine production confined to PEMT methyl transfer. [1] [19]

3. Levers

Dietary methyl-resource provision is supported; no intake optimisation protocol is established.

4. Mechanistic Basis

Summary

PC formation has two distinct routes. Hepatic PEMT uses SAMe to methylate phosphatidylethanolamine. The Kennedy pathway builds PC using choline, CTP and diacylglycerol; uridine can contribute to its nucleotide supply. Neither route alone establishes brain DHA uptake or cognitive benefit. [1] [19]

5. BRS Pathways and Connections

5.1 BRS Pathways

BRS2-FM1-PM1 — Folate/B12-Dependent Homocysteine Remethylation ↓ BRS2-FM3-PM7 — Phosphatidylcholine Formation ↓ BRS1-FM3-PM7 — Neuronal Membrane DHA Incorporation

5.2 Cross-BRS Mechanism Relationships

Primary Mechanisms in other Biological Regulatory Systems that directly interact with, constrain or support this mechanism.

  • BRS1-FM3-PM7 — Neuronal Membrane DHA Incorporation — Hepatic methylation produces phosphatidylcholine from phosphatidylethanolamine. Later blood–brain transport of DHA and its uptake into neuronal membranes are a downstream delivery step, not this methyl transfer.

5.3 Local BRS Mechanism Relationships

Related Primary Mechanisms within the same Biological Regulatory System that collectively support the integrated biological function.

  • BRS2-FM1-PM2 — Betaine/BHMT Remethylation — Choline-derived betaine contributes methyl groups through homocysteine remethylation to methionine and subsequent SAMe synthesis. This upstream chain supports methyl-group provision to PEMT (SF7 — Dietary choline forms supply methyl groups to the PEMT route).

  • BRS2-FM1-PM3 - SAMe Synthesis — Methionine adenosyltransferase produces the S-adenosylmethionine consumed in each phosphatidylethanolamine methylation. That is upstream methyl-donor supply, not the methylation reaction itself.

7. Phenome Connections

These mappings are translational relationships, not single-mechanism outcome claims. Phenomes are emergent functional patterns supported by multiple interacting PMs across the BRAIN Framework.
