BRS-X(ECS)

BRS-X-ECS-1-2 - Omega-3-Derived Endocannabinoidome Signalling

1. Mission & Overview

Mission

Sustain omega-3-derived ethanolamide production so endocannabinoidome neuromodulation stays supplied from dietary fat status.

Intervention Dominance: Diet-Dominant — Dietary Requirements

The established influence is bounded to the measured endpoint and exposure context.

Overview

EPA and DHA can supply the fatty-acid portions of omega-3-derived signalling lipids. DHA conversion into synaptamide has been traced in neuronal cells, while controlled exposures change related ethanolamides in human plasma. This gives a specific precursor-supply role to long-chain omega-3 inputs. It does not make all omega-3 molecules interchangeable: ALA conversion, delivery form, tissue metabolism and degradation affect the route. Neural production and clinical responses must be distinguished from circulating lipid measurements. Kevala et al. (2020) [4]; Pu et al. (2016) [5]; Yang et al. (2019) [6]

  • Biological Relevance: Both exposures supplied N-docosahexaenoyl-PE and synaptamide; equimolar free DHA generated more synaptamide. Kevala et al. (2020) [4]
  • Implementation Notes: Use the bounded precursor-supply relationship below; its context and limitations travel with the disclosure.

2. Primary Biological Effects

  • Both exposures supplied N-docosahexaenoyl-PE and synaptamide; equimolar free DHA generated more synaptamide. Kevala et al. (2020) [4]
  • DHA-enriched oil increased its corresponding plasma ethanolamide. EPA-containing arms had higher plasma EPEA than DHA alone; combined exposure also changed DHEA. Pu et al. (2016) [5]; Yang et al. (2019) [6]

4. Mechanistic Basis

Summary

Both exposures supplied N-docosahexaenoyl-PE and synaptamide; equimolar free DHA generated more synaptamide. Kevala et al. (2020) [4]

Mechanism boundary: ALA-to-EPA/DHA-to-neural ethanolamide supply was not established as a PM-specific dietary relationship; no automatic ALA admission.

Scientific Findings and their source assessments are available in Evidence & Audit.

5. BRS Pathways and Connections

5.1 BRS Pathways

  • None listed

5.2 Cross-BRS Mechanism Relationships

Primary Mechanisms in other Biological Regulatory Systems that directly interact with, constrain or support this mechanism.

5.3 Local BRS Mechanism Relationships

Related Primary Mechanisms within the same Biological Regulatory System that collectively support the integrated biological function.

Phenome assessment boundary: Existing ratings and their original source records are retained pending outcome reassessment. The new mechanism and lever Findings do not validate or raise those ratings.

7. Phenome Connections

These mappings are translational relationships, not single-mechanism outcome claims. Phenomes are emergent functional patterns supported by multiple interacting PMs across the BRAIN Framework.
