BRS3-1-1 - NF-kB Signalling Regulation
(Turning Down Overactive Inflammatory Switches)
1. Mission & Overview
Mission
Regulate NF-κB transcriptional tone so pro-inflammatory gene programmes do not fire excessively.
Intervention Dominance: Not established — No principal intervention route established
Precursor tracing measures red-cell glutathione; NF-κB redox studies use experimental cells. Their combination does not establish human dietary intervention responsiveness. No route is ranked above another, because none is established.
Overview
NF-κB helps cells translate immune signals into inflammatory gene programmes. Its activation and DNA binding are regulated at several steps, including inhibitor phosphorylation and glutathione-dependent redox control. [7] [11] [12] Keeping this response appropriate is the biological aim; suppressing every response is not an established health benefit.
- Benefits: This mechanism governs inflammatory signalling rather than a proven nutrient treatment or clinical outcome. The reviewed experiments establish regulation of activation and DNA binding. [7] [11]
- Implementation Notes: Cysteine and glycine have supported upstream roles through glutathione. Combined supplementation increased red-cell glutathione synthesis in older adults, but did not measure NF-κB or establish a food dose. [13]
- Biological Relevance: Glutathione provision connects this PM to redox biology; NF-κB regulation remains distinct from glutathione synthesis and downstream cytokine-network outputs. [11] [12]
2. Primary Biological Effects
NF-κB activation, DNA binding and redox-sensitive inflammatory gene regulation
3. Intervention Levers
No Direct or Derived Dietary Requirement is currently established for regulation of NF-κB transcriptional tone.
- Magnesium ions (Mg²⁺)
- Tested fibre/fruit-rich mixed meal versus the high-fat/high-carbohydrate comparison
No evidence-qualified System Optimisation Practices are currently established for this mechanism.
No lifestyle lever is established. Sleep, activity, and meal timing were not shown to change NF-κB transcriptional tone.
4. Mechanistic Basis
Summary
When inflammatory transcriptional programmes remain engaged, downstream cytokine and immune-signalling tone escalates. NF-κB is a principal upstream node through which those programmes are expressed within BRS3(FM1) - Anti-Inflammatory Signalling Tone.
(NF-κB as an inflammatory transcriptional node)
Mercurio et al. (1997) identified IKK-1 and IKK-2 as the kinases that phosphorylate the inhibitor proteins holding NF-κB, and showed that IKK-2 is required for NF-κB activation in the cell systems tested. [7] That kinase step permits activation; it is not transcription itself. The cytokine messengers that follow it are a later step — see PM7.
(Signalling pressure and pattern context)
Glutathione-dependent redox regulation acts on IKK activity and NF-κB DNA binding. [11] [12] Redox state can inhibit as well as permit activation, so more precursor supply cannot be assumed to suppress inflammation. The attached dietary-pattern studies do not independently establish a PM-specific intervention. Adams (2001) reviews ATP-dependent protein-kinase chemistry, including the usual metal partner of that reaction. [8] That chemistry is not a dietary magnesium result.
(Boundaries of the mechanism)
Downstream cytokine-network expression is handled by BRS3-FM3-PM7 - Cytokine Network Modulation. Gut-derived endotoxin load is represented by BRS3-FM1-PM2 - Gut-Derived Inflammatory Signalling. EPA/DHA as specialised pro-resolving mediator substrates, and review claims that SPMs downregulate NF-κB, belong to BRS3-FM3-PM8 - Eicosanoid / SPM Balance. This PM does not currently carry an evidence-qualified EPA/DHA dietary requirement or System Optimisation Practice for NF-κB transcriptional tone. Endogenous antioxidant induction and net redox balance belong to BRS3-FM2-PM3 - Nrf2-Mediated Cellular Defence Regulation and BRS3-FM2-PM4 - ROS Generation vs Clearance Balance.
(Integration within BRS3)
This PM anchors transcriptional inflammatory tone within BRS3(FM1). Gut-barrier context imported through BRS5-FM1-PM1 - Gut Barrier / Tight Junction Integrity conditions endotoxin-linked pressure on this pathway.
4.1 Scientific Findings (3)
Scientific Findings and their assessments are available in Evidence & Audit.
Summary
IKK experiments establish an activation gate, while redox studies demonstrate glutathione-dependent regulation of kinase activity and DNA binding. Human precursor tracing supports glutathione provision, but does not measure NF-κB. Together these findings support a bounded upstream supply relationship, not a proven dietary anti-inflammatory effect.
5. BRS Pathways and Connections
5.1 BRS Pathways
- None listed
5.2 Cross-BRS Mechanism Relationships
Primary Mechanisms in other Biological Regulatory Systems that directly interact with, constrain or support this mechanism.
- BRS5-FM1-PM1 — Gut Barrier / Tight Junction Integrity — A compromised tight junction lets lipopolysaccharide cross. That is inflammatory pressure on this transcription factor, not the IKK phosphorylation step.
- BRS5(FM1) — Gut Barrier Integrity and Immune Interface — The gut–immune interface sets how much microbial product reaches systemic signalling. That is upstream context for this transcription step.
- BRS6-FM1-PM2 — Glycaemic Variability Regulation — Postprandial glycaemic swings are cited as inflammatory context around this pathway. They are not the IKK phosphorylation reaction.
5.3 Local BRS Mechanism Relationships
Related Primary Mechanisms within the same Biological Regulatory System that collectively support the integrated biological function.
- BRS3-FM1-PM2 - Gut-Derived Inflammatory Signalling — Gut-derived signalling is the barrier-entry step that can raise the inflammatory load this transcription factor responds to.
- BRS3-FM3-PM8 - Eicosanoid / SPM Balance — Eicosanoid and resolving-mediator balance is a later lipid-signal step. It is not the IKK release of NF-κB.
7. Phenome Connections
These mappings are translational relationships, not single-mechanism outcome claims. Phenomes are emergent functional patterns supported by multiple interacting PMs across the BRAIN Framework.
Each Phenome relationship has two independent ratings. They are not combined or averaged.

- Evidence Confidence: Low–Medium
- Rationale: LPS-linked neurotransmission disruption intersects depressive mood biology in translational reviews; pro-inflammatory transcriptional tone may modulate this context without direct anhedonia-outcome claims.
Stage 2A/2B dietary-target assessment — 23 October 2026
ENRICHED. NF-kB transcriptional activation, distinguished from upstream redox supply.
Admit the mixed-meal comparison only as a bounded SOP; the study directly measured mononuclear NF-kB DNA binding. Do not attribute the response to isolated fibre, orange juice or polyphenols, or treat upstream GSH as direct evidence of transcriptional regulation.
New implementation: 0 dietary-input admissions (0 completed Unset candidates), 1 practices, 0 biochemical participants, 1 unscored Findings. Existing accepted inputs, source assessments, mission, KCs, Phenome relationships and scores are retained.
Source-to-claim boundary: Assessment source. Stop after adjudicating the stated material candidate and tracing supported delivery. Keep uncertain specificity, formulation, anatomical or measurement links explicit; no negative evidence is inferred from incomplete retrieval.
