BRS-X-Hormones-1-3 - Progesterone-Supportive Microbial Metabolism
1. Mission & Overview
Mission
Sustain microbial contributions to progesterone-related hormonal stability through gut ecosystem function.
Intervention Dominance: Diet-Dominant — No principal intervention route established
The inherited intervention qualification is not confirmed by a mission-specific dietary/practice effect in this assessment.
Overview
Gut bacteria can convert a host-derived biliary steroid into progestin metabolites. Defined culture and mouse experiments identify hydrogen as an important resource for this conversion. This gives the page a concrete microbial process to assess, but it does not yet establish maintenance of circulating progesterone or hormonal stability. The distinction matters for implementation: generic fibre intake or a named bacterium cannot be presented as a hormone-supporting requirement without evidence connecting that exposure to the conversion and the relevant physiological endpoint. McCurry et al. (2024) [2]
- Biological Relevance: Hydrogen supported conversion of 3alpha,5alpha-THDOC into tetrahydroprogesterone products; human stool observations complemented the experiments. McCurry et al. (2024) [2]

4. Mechanistic Basis
Summary
Hydrogen supported conversion of 3alpha,5alpha-THDOC into tetrahydroprogesterone products; human stool observations complemented the experiments. McCurry et al. (2024) [2]
Mechanism boundary: The demonstrated process is progestin production, not progesterone stability. Canonical fermentable-fibre members to converter-accessible hydrogen and the hormonal endpoint need separate adjudication.

5.2 Cross-BRS Mechanism Relationships
Primary Mechanisms in other Biological Regulatory Systems that directly interact with, constrain or support this mechanism.
- BRS-X(Hormones-PM2) — Estrobolome Regulation — Assesses microbial estrogen deconjugation; systemic recycling requires its own endpoint evidence.
- BRS-X(Hormones-PM4) — Metabolic-Reproductive Hormone Integration — Assesses insulin–steroid integration; microbial hormone conversion is not the same biological job.
- BRS1(FM1) — Monoaminergic Function — Candidate contextual connection retained for review; an exposure-specific resource or signalling relationship is not yet characterised.
- BRS5-FM2-PM5 — SCFA Production & Signalling — Candidate contextual connection retained for review; an exposure-specific resource or signalling relationship is not yet characterised.
- BRS5(FM1) — Gut Barrier Integrity & Immune Interface — Candidate contextual connection retained for review; an exposure-specific resource or signalling relationship is not yet characterised.
- BRS6(FM1) — Glycaemic–Insulin Stability & Cognitive Energy Availability — Insulin availability provides metabolic context for steroid synthesis; a clinical response requires the population-specific evidence retained here.
5.3 Local BRS Mechanism Relationships
Related Primary Mechanisms within the same Biological Regulatory System that collectively support the integrated biological function.
- BRS-X(Hormones-PM2) — Estrobolome Regulation — Assesses microbial estrogen deconjugation; systemic recycling requires its own endpoint evidence.
- BRS-X(Hormones-PM4) — Metabolic-Reproductive Hormone Integration — Assesses insulin–steroid integration; microbial hormone conversion is not the same biological job.
Phenome assessment boundary: Existing ratings and their original source records are retained pending outcome reassessment. The new mechanism and lever Findings do not validate or raise those ratings.
7. Phenome Connections
These mappings are translational relationships, not single-mechanism outcome claims. Phenomes are emergent functional patterns supported by multiple interacting PMs across the BRAIN Framework.
Each Phenome relationship has two independent ratings. They are not combined or averaged.
Biology → Phenome Relationship Strength
How direct and important is this biological mechanism expected to be to the Phenome within the BRAIN model?
Evidence Confidence
How strongly does the adjudicated evidence support this particular biology → Phenome relationship?
- Biology → Phenome Relationship Strength: Low
- Evidence Confidence: Low
- Rationale: Butyrate-producing taxa may support endocrine and inflammatory contexts relevant to progesterone balance, but direct ADHD outcome evidence is limited.
- Biology → Phenome Relationship Strength: Low
- Evidence Confidence: Low
- Rationale: Microbial SCFA context may indirectly influence stress-responsive endocrine-inflammatory tone intersecting progesterone-related stability.
- Biology → Phenome Relationship Strength: Low
- Evidence Confidence: Low
- Rationale: Progesterone-linked calming tone may intersect with gut-derived inflammatory and SCFA signalling, but evidence for direct dietary single-mechanism effects remains limited.
