BRS3 — Inflammation & Oxidative Stress

BRS3-3-8 - Eicosanoid / SPM Balance

(Shifting from Pro-Inflammatory to Calming Pro-Resolving Lipid Signals)

1. Mission & Overview + Dietary Levers

Mission

Shift lipid mediator balance from pro-inflammatory eicosanoids toward pro-resolving signals that terminate inflammation.

Short-term n-3 fatty acid supplementation raised some circulating pro-resolving mediators in healthy adults. Not every mediator rose, and this is not a disease-resolution outcome.

Intervention Dominance: Diet-Dominant — Dietary Requirements

Not every specialised pro-resolving mediator rose. Aspirin added no further rise. This is not a disease-resolution outcome. The rise is in healthy adults and does not include every mediator.

Overview

Resolvins and protectins are made from EPA and DHA, and they help inflammation finish rather than only blocking it. Serhan and Petasis (2011) describe that pathway. [1] Barden et al. (2014) found that 2.4 g a day of n-3 fatty acids for five days raised RvE1 and several pathway markers in healthy adults. [9] Not every mediator rose.

  • Benefits: These mediators support the finishing of inflammation, which is a different job from blocking the signal at the start. [1]
  • Implementation Notes: EPA and DHA are the substrates. [1] The intake that raised some mediators was 2.4 g a day for five days in healthy adults, not a disease-resolution dose. [9]
  • Biological Relevance: Cytokine intensity is a later messenger readout — see PM7.

2. Primary Biological Effects

↑ pro-resolving mediator bias; ↓ pro-inflammatory eicosanoid dominance

3. Intervention Levers

4. Mechanistic Basis

Summary

Inflammation termination depends on lipid mediators that actively resolve rather than merely suppress immune activity. EPA and DHA substrate availability shapes whether signalling bias moves toward specialized pro-resolving mediators within BRS3(FM3) - Inflammation Resolution Capacity.

5. BRS Pathways and Connections

5.1 BRS Pathways

  • None listed

5.2 Cross-BRS Mechanism Relationships

Primary Mechanisms in other Biological Regulatory Systems that directly interact with, constrain or support this mechanism.

5.3 Local BRS Mechanism Relationships

Related Primary Mechanisms within the same Biological Regulatory System that collectively support the integrated biological function.

7. Phenome Connections

These mappings are translational relationships, not single-mechanism outcome claims. Phenomes are emergent functional patterns supported by multiple interacting PMs across the BRAIN Framework.
